Spravato® is a prescription nasal spray medication, used along with an antidepressant taken by mouth to treat:

• Adults with treatment-resistant depression (TRD)

• Depressive symptoms in adults with major depressive disorder (MDD) with suicidal thoughts or actions

Spravato® is the first FDA-approved nasal spray medication, taken with an oral antidepressant, for treatment-resistant depression in adults. 

​

If you’ve tried two or more antidepressants without sufficient relief, talk to your healthcare provider to see if you may have treatment-resistant depression.

​

Spravato® targets the N-methyl-Daspartate (NMDA) receptor and is believed to work differently than currently available oral antidepressants.

How Spravato® Works

SPRAVATO® is different from other anti depressant medications because it acts on the glutamate pathway.

Clinical study results suggest that glutamate transmission is abnormally regulated in multiple areas of the brain of individuals with depression.

​

Spravato® works by antagonizing the NMDA*** receptor and activating the AMPA* receptor, which leads to downstream release of brain-derived neurotrophic factor (BDNF**) and activation of kinases.

​

BDNF expression and downstream signaling are thought to enhance neurogenesis and mediation of synaptic excitability and plasticity.

 

Spravato® may help improve symptoms of depression in individuals who have not responded to other treatments.

​

It is important to note that the relationship between this pathway and the clinical effects of depression have not been established.

AMPA* = α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid.

BDNF** = A brain-derived neurotrophic factor.

NMDA*** = N-methyl D-aspartate - A brain receptor involved in memory and learning.
